Dandruff is caused by lipid (fat) autoxidation damage to scalp cells, resulting in their proliferation and then falling off. Rancid, musty hair and scalp odors are also caused by autoxidation of lipids in the scalp and on the hair. It can be effectively controlled by shampoos containing antioxidants, such as selenium sulfide (found in Selsun Blue® shampoo) and zinc
pyrithione (the latter is a zinc-sulfur compound found in Head and Shoulders® shampoo). Before taking supplementary antioxidants, one of us had a lot of dandruff and some hair odor, which he kept under control but could not entirely eliminate with Head and Shoulders® shampoo. Now he does not have to use antidandruff shampoos. But if he goes off his antioxidant supplements, the dandruff and hair odor
begin to return in about two weeks. A friend of ours has reported that selenium sulfide shampoo reduced his hair loss.
Male pattern balding is a convenient readily accessible aging clock. One needs neither laboratory instruments nor decades to observe the impact of experimental manipulations on this aging clock. One can affect it with topical treatments having negligible systemic effect. And if one inadvertently makes this-clock tick faster rather than slower, the results are not likely to be fatal (which they could be with the hypothalamic-pituitary clocks) or even permanent. Of course, only some of the things that affect this aging clock will affect aging clocks in general. Topical low-diffusability sex hormones applied with the proper solvent definitely do affect
this aging clock but should not be expected to affect aging clocks in general. Both antioxidants and polysorbates 80 and 60 may have more general aging clock effects, since both classes of compounds can cause renormalization of some very early precancerous tissue developmental alterations, and
since some aging clocks, such as those in the testosterone-producing Leydig cells in the testes and in the dopaminergic hypothalamic-pituitary nerve tracts, involve lipofuscin-accumulation-induced shutdown which could be a type of “mutation monitor” measuring free radical damage.
Finally, there are compounds such as minoxidil (Upjohn’s Loniten®), diazoxide (Schering’s Hyperstat®), and clonidine (Boehringer Ingelheim’s Catapres®), all three of which are approved by the FDA for the control of severe high blood pressure. These promote tissue repair where repair does not normally occur; we think they may affect genetic expression. The first two drugs have hirsutism (excess hair growth) as side effects, and the third caused an improvement in memory and learning in some patients with Korsakoff’s psychosis (alcoholic brain damage). Of the three drugs, only minoxidil is FDA approved as a hair restorer.
The future undoubtedly holds many more compounds which
selectively stimulate tissue regeneration.
